    Isolation and Identification of microorganism in Mango with Anthracnose disease

    Abstract
    Mango (Mangifera Indica L.) anthracnose is a disease that mostly occur in plant. It usually attack on fruit, leaf, stem and root. The aim of this study is to isolate and identify the microorganisms from mango with anthracnose disease. In this study, the rotten mango with anthracnose disease symptom are used as a sample. The black spot on the fruit are cut and put on Potato Dextrose Agar (PDA). A week of incubation at 27˚C are resulted in grow of microorganism on the PDA plate. The samples were undergo streak plate technique to obtain a pure culture. The selected microorganism were identified by classified them as Isolate 1, Isolate 2, Isolate 3 and Isolate 4. DNA extraction are proceed after that to get a genomic DNA. The molecular identification of the DNA by polymerase chain reaction are by using 16s rRNA with primer 27F and 1492R as target sequence. The PCR product of approximately 1 500 bp of these isolates were sent for sequencing. Chromas Lite is a software used to read the DNA sequence. The obtained sequences were used to identify species and to construct phylogenetic tree then compared the sequences in GenBank using online software known as BLAST and Clustal Omega. All of those four isolate were identified as Pseudomonas sp. These microorganism can be utilized in future since it can be a biologically agent in control anthracnose disease.
