    Abstract
    Spider silks possess an extraordinary combination of high strength and elasticity that is made up of unique Protein spidroins. The aim of this study is to investigate the spider silk processing from the webs of Crossopriza Lyoni for silk hydrogel fabrication. 20 mg of pre-treated spider web was subjected to 20 ml of 0.5 % extraction solution contain Sodium Dedocyl sulfate,water and ethanol for extraction at a selected range of temperature, SDS concentration and ethanol ratio. The significant parameters were selected by using statistical approach of Full Factorial Design through screening process. The screened parameter was subjected for optimization by using Central Composite Design for statistical approach. One-way anova was used to determine the significant of the quadratic model using its p-value of the interaction affecting the spidroin extraction. Factor affecting the extraction condition were statistically optimized using Response Surface Methodology (RSM). for maximal recovery of spidroins. The optimum extraction condition obtained at 500C temperature and 6 ratio of ethanol to water underwent the validation process to check the accuracy of the model equation. The extraction process was continued with fabrication of the Hydrogel from silk spidroin solution. The extraction is using specific condition derive from the optimization process whereby the highly viscous protein solution were mold and became hydrogel.
